Can a GAGome score predict who will develop cancer over the next five years, among people who currently have none?

Screening is offered on the basis of risk, estimated from age, smoking history and family history. People those models place below the threshold are not screened.

Design: Prospectively planned retrospective case-cohort validation study in adults aged 35–80, asymptomatic at sampling with no recent cancer history. Biospecimens and linked outcomes come from population-based biobank cohorts, with diagnoses confirmed by biobank records or cancer registry linkage. The score and cut-off were locked in a development sub-study before independent validation.

Status: Development complete and the score locked. A manuscript reporting that work is under review.

Lifelines Cohort Study is a multi-disciplinary prospective population-based cohort study examining in a unique three-generation design the health and health-related behaviours of 167,729 persons living in the North of the Netherlands. It employs a brood range of investigative procedures in assessing the biomedical, socio-demographic, behavioural, physical and psychological factors which contribute to the health and disease of the general population, with a special focus on multi-morbidity and complex genetics.

The Lifelines initiative has been made possible by subsidy from the Dutch Ministry of Health, Welfare and Sport, the Dutch Ministry of Economie Affairs, the University Medical Center Groningen (UMCG), Groningen University and the Provinces in the North of the Netherlands (Drenthe, Friesland, Groningen).

LEVANTIS-0087A is registered at ClinicalTrials.gov (NCT05235009).
